Abstract
Recently, significant accomplishments have been made in the development of new treatments for spinal cord injury (SCI) from basic science research through clinical application. The far more challenging goal of restoring bladder function to improve the quality of life among SCI patients has been largely underachieved. The spinal cord segments below the injured level undergo remodeling to adapt to changes in functional outcomes. In this proposal, we will use both subacute and chronic SCI pre-clinical rodent models to test the efficacy of proposed interventions. Particularly, chronic SCI is the condition faced by the majority of the SCI population, and there are still no effective treatments available for these patients. We will also use a contusive SCI experimental model, which closely mimics the injury of many patients with SCI. We propose to use the combination of both pharmacological intervention and rehabilitation approaches to maximize the improvement of both bladder and sensory functions following SCI. The drug (NLX112) that will be used is currently a Phase II-ready drug intended for the treatment of L-DOPA-induced dyskinesia, which is a side-effect of Parkinson's disease treatment and rehabilitation that is routinely observed in medical practice. Therefore, success in this project can be translated toward clinical application in patients with SCI to address autonomic dysfunctions. ( chn:wdg)
